I have Windows 10 on my PC, and the game used to run. The other day--after I exited out of the game because the screen was glitching--I tried to run it again, and nothing happened. Today, I'm having the same problem, along with the fact that I can't uninstall and then reinstall it. I've tried several of the compatibility modes, of which none worked.

@IamveryClassy The Origin issue may or may not be related, but you certainly need to get Origin to work before troubleshooting the game, or you might end up wasting a lot of time. For Origin, don't use any compatibility mode—it's not necessary and is often counterproductive. Start by clearing its cache:

If it still won't update properly, clear its cache again, then uninstall "the hard way." This shouldn't affect any games you have installed.

Instead of using the ThinSetup, download the full installer. Be sure to run it as an admin: right-click on the installer and select "Run as administrator." This is a direct download link:

Let me know if Origin still doesn't run properly. If it does, you can try Sims 3 again, although don't use any compatibility mode for it either. (The game runs just fine in Windows 10.) If TS3 doesn't work, please let me know exactly what happens when you press play in Origin. Please also run a dxdiag and attach it to a post.
