So you get the same error at installation? (Failed to initialize the physics system)
Did you tried to install the PhysX software in this way?
- Download the PhysX software from here.
- De-install the PhysX software from the Windows control panel > reboot.
- Run the "PhysX-9.16.0318-SystemSoftware.exe" as Administrator > reboot > test

I did it now again as you said (uninstall physx, reboot, install newest downloaded physx driver as administrator, reboot) and i get the same error message when i try to launch the game from the origin client.

Ok, that is strange.
Is ME3 the only games with problems?
Please go to your ">drive<:\Program Files (x86)\Origin Games\Mass Effect 2\__Installer\physx\redist" folder and run the "PhysX-9.12.0613-SystemSoftware.msi" > remove > reboot > go back and run the "PhysX-9.12.0613-SystemSoftware.msi" again.
If that don't helps, run the Windows System File Checker Sfc /scannnow and post the concluding message.

Yes, i have no problem with any other games.
And yes, i already tried with the physx version that can be found in the game installer directory.
(anyway its ME3, and PhysX-9.11.1107-SystemSoftware.msi )
sfc done.
Windows Resource Protection did not find any integrity violations.
any ideas?

@holger1405 now i tried Mirrors Edge with full physx support, and runs perfectly.

Not many to be honest.
I assume the repair function is not available yet? (in Origin > Game Library > right click ME3 > Repair)
If not I would try to reinstall the game running Origin as Administrator.

After two weeks of misery, the final solution: windows re-install. Now everything's fine
