Ok, then try it this way.
- If that don't works, download the NVIDIA PhysX software from here.
- deinstall the NVIDIA PhysX software from the Windows control panel > restart > install the downloaded NVIDIA PhysX software > restart > test.

OK did that and launched Origin which is now auto updating DLC which had already installed so can't select play yet. Hopefully only 5 to 10 minutes

This was excellent advice, Holger, running the .msi from the game folder.
This was an odd thing, but it worked and I am back in-game in minutes

Hi guys!
I've tried everything here written, but it still doesn't work.
Uninstall physx, restart, install from game folder, restart, try.
Uninstall, restart, install the latest from official nvidia download, restart.
Anyone any other advice?

Please create a DxDiag in text file format, also go to your Reliability monitor (Press the windows key and "R" at the same time and copy or type "perfmon /rel" (without the quotes) in > ENTER and look if it has ME3 entries.
If yes double click at the last one, copy it to the clipboard, (Bottom left corner) save it to a text file and load it up together with the DxDiag. You can do that with the "choose file" button at the bottom right corner of the reply window.
